Belize hotels are generally not like Mexico hotels with huge pools, swim up bars, all you can eat buffets and swarms of people. Instead you will see more medium sized hotels with natural beauty all around them. That being said there are many very luxury hotels on Ambergis Caye, in the Placencia area and a handful of private island resorts.
Being such a small country it is possible to visit all the major areas in Belize over a couple of weeks but it is best to choose a home base. What you should decide is do you want to be close to the barrier reef, the jungle, the ruins, fishing etc.. All luxury hotels will be situated close to all the above but some will be better suited for say fishing or jungle tours.
For example if you choose to stay out on the turneffe Atoll then you may not be spending much time exploring the jungle since it is many miles out to sea. But the fishing would be fantastic as would the diving and snorkeling.
These are some things to keep in mind when choosing a luxury belize hotel.
